On linux only one program at a time can access MTP devices. Get your filemanager/desktop environmnet to not auto mount hte device and it will work in calibre.

MTP type USB on Linux only support one connection / application.
On some Linux or desktops you can disable automount, then Calibre can connect.
On some MTP android you can Eject/unmount on the desktop and then Calibre can connect.

I use Mate desktop on Mint, so before USB connection I go to to System Monitor and stop (not kill as that auto-respawns) Processes -> gvfs-mtp-volume-monitor, right click and Stop.

Because while disabling "automount" works in Cinnamon, it is ignored by Mint.

Then connect tablet/phone.
Calibre will connect.
After eject in Calibre and unpluging you can "continue" the stopped process if you need to use the file manager with an MTP device.
