hello and kindle question

Hi all. I am new to e readers and seem to need a little advice. Thanks!

I just purchased my first e reader and was advised to download calibre for format conversions. Things seemed to go fine last night and I transferred some books from calibre to the Kindle. Now today I cannot get calibre to see my device??

I have tried several USB ports on my iMac running 10.5.8. Tried with Kindle off then on, on then off etc. NO go. I can see the Kindle disc on the desktop when on USB, and it works fine for wireless downloads from Amazon. Any advice?
