I was use squashfs it in my Linux LiveCD before.
But can you explain more about its benefits wih Kindle?

i build a debian chroot env with a ext3 loop file ,the size is about 450M, when switch to squashfs ,it's about 150M ,so I chose squashfs

I compile a brower named arora based on qt for kindle dx us to view local html file of qt4.7.1 doc .

squashfs is very space efficient ,see below, so it's can also to use with original reader. but it's readonly filesystem. so the reading position restore may not work.
