It does not necessarily have to be on your c:\ drive.
It just needs to be on a local drive... ie a hard drive that is inside the pc you are using.
My calibre library is on my e:\ drive which is a local drive along with f:\ and w:\ (mirror drive)

In my limited testing, SQLite is very sensitive to network and timing issues. As suggested by multiple people, using a NAS or cloud storage for a live calibre library is strongly not recommended. Do a one way sync to NAS or cloud storage as a backup (do not use GoogleDrive!), use them for backing up your files, just do not access your working library on them.

That uses Mass Storage. Obviously USB Networking or MTP won't work, but they are usually only "gadgets" with storage, not dedicated storage devices.

Also the drive ideally should use the native file system, not FAT32 or exFAT.

That was what resolved it. I placed the library on an external HD stuck on the USB. It worked.
