Send to Device: a book in two formats

Assume a book in a calibre library in both epub and azw3 formats.

With either a Kindle connected or a Kobo, what happens during the send?

The list of book formats that will be sent to a device is listed in the driver configuration in preferred order. So a Kobo would receive a format from its list and a Kindle would receive a format from its list. If I remember correctly, if you select a book that does have a format enabled in the device list, a format will be selected, converted and sent.

One friend of mine uses both Kobo and Kindle devices but keeps only ePubs in their library. When a Kindle is connected, the ePub will be converted to AZW3/KF8 before sending.

And in converse, if the book(s) is azw3 in the library and a Kobo is connected, calibre converts it on the fly with send?

Yes. Personally, I prefer to have ePubs in my library and as a source format for other formats.

Basically, there are more editors for ePub format ebooks though my personal preferences are Sigil and calibre's ebook-editor for some specialized functions.

There is no equivalent for epubcheck or epubveri for other formats to allow error checking. Most publishers want an ePub as an input document and that document must pass epubcheck before they will accept it.
