Calibre has, to the best of my knowledge, not been tested with the Big Sur beta
so the information that you are running a pre-release OS is an fairly important data point and would have affected the answers you received.

Perhaps a response such as "did you try going into Security and Privacy, Privacy, Full Disk Access and adding calibre to the list of apps there" as a test?

I've read about enough people running OS X beta with issues with Readers. So IMHO, if you have a Reader, do not run any OS X beta.

WHy are you running a Big Sur beta is you don't understand how to use it? You would be better off dropping Big Sur for Catalina. One problem it could be is that QTWebKit is not fully Big Sur compatible.

Bleeding edge is for the adventurous and those that recognize there be beta issues happening. Even Gamma level is a risk of odd things happening.
