\U is not a supported operator by the regex engine calibre uses.

Possibly you could add operator icons to the Text Formatting Toolbar to UPPER CASE lower case Proper Case First letter only all selected text not within < > pairs?? Potential workaround |

As I've said before, there will be a function mode for search and replace, that will allow doing this and many, much more powerful operations.
