Running Kindle on chromebook via Linux.
Although the kindle is connected to chromebook as evidenced in folders when I run the debug it identifies that it is not connected (see below)
This is further evidenced that when I try to transfer books to kindle the transfer to main memory etc is greyed out.

You might try to update 5.30.1 (a hot fixed for 5.30)

Your chromebook is not seeing *any* usb devices, probably because its usb subsytem is disabled. See the line

The Chromebook Unix system does not provide the support required for usb device detection.

What I don't understand is why the system has operated previously without any problems? The none recognition is a new issue

On a Chromebook I've used with Crostini, a ChromeOS update broke it's USB subsystem. From what little I was able to find out, it was broken due to a security update.

Sadly I'm not technically minded. Is there anything I can do to rectify the problem?

Stop using that OS, since the feature YOU want has not been fixed.

There is nothing a 3rd party developer can do to make your Hardware turn on.
(that in itself would be a Security flaw)

I've just connected an external hard drive via usb and am able to write to and transfer data/folders. Doesn't this infer its a problem with Calibre?

Can your chromebook SEE your device? The issue might be the type of device (reader, is not seen as a disc).

No, calibre runs in the linux subsystem, chromeos exposes usb devices at a different layer.
