Sorry, I must have missed seeing that.
Let me recap my understanding of what you're telling us:
In Windows Safe mode you can read an epub that is in a calibre library, but in Windows "Normal" mode you can't read the same book in the same library - even after disabling all Start Up tasks
and restarting Windows.
Did you use Safe Mode Minimal?

Whoa, that suggests it's the data!
What did you do to the book on the desktop that won't open, that you did
not do with the book on the desktop that will open - e.g. convert, edit, embed metadata etc etc.
