Are you using an old version of Calibre?

The only time I recall seeing that error it was to do with directory locking by window's file explorer conflicting with Calibre.

Newer versions of Calibre report this in a way FFF catches it and offers you a chance to close file explorer before trying again.

Otherwise, I'd need a debug log to even start looking at it.
