Did you try the suggestion in the thread in this forum called Calibre not launching. NUL file missing in windows or the oldest thread I could find when I did a forum search on "NUL file missing in windows" (the quotes are needed) which is Calibre Won't Run from Jan.,2012. The advice given at Nuls R Us was intended for Windows XP and does not work on later Windows versions.

You could even use a Google search for "NUL file missing in windows" site:mobileread.com to get even more search results from multiple MobileRead forums.
