Looking at the error, that is saying that when calibre tries to access something on the tablet, it is failing. It seems to be the card in the tablet. Have you tried to use Windows to access the card when the tablet is connected? Have you tried removing the card and seeing if calibre can see the tablet? Have you tested the card directly in the computer or some other device? Maybe it is failing. Or try an other card in the tablet.

Thanks for the detailed reply. Im off on holiday tomorrow so may resort to a paper-backs for now.
Unfortunately I don't have another PC to try on. Are you thinking its a firewall problem - I hadn't considered that.
Calibre is NOT able to access either the built-in memory of the S2 tablet or the card. When I try to access either, this is when the error is thrown up. I can access both the tablet structure and see/access book files on the card. Its when I try to add a new book either to the main memory or the sd card that the problem arises. Furthermore I can still read books from the card - dont think I could do that if the card was corrupted??
I have a screen-shot from my PC but cant seem to attach it- will try again as an additional post
