Yes, there is a good reason not to do that. Because it is unlikely to be calibre causing the problem. A better thing to do would be to go to another PC and try again. If it fails, then it is the device. If it doesn't fail, it is probably the first PC.
Looking at the error, that is saying that when calibre tries to access something on the tablet, it is failing. It seems to be the card in the tablet. Have you tried to use Windows to access the card when the tablet is connected? Have you tried removing the card and seeing if calibre can see the tablet? Have you tested the card directly in the computer or some other device? Maybe it is failing. Or try an other card in the tablet.
