DO NOT willy-nilly try your own repair as you can destroy/invalidate the metadata backup files in each folder (metadata.opf) if you do this incorrectly.

Wait for further advice from the developers. (and you can read the FAQ pages about similar topic)

In the meantime:
1) Reboot
2) disable your antivirus/firewall ( Disconnect you network cable/turn off WiFi while this is being done)
3)Start Calibre. Success? an update of #2 is probably where you need to do work.
