The only difference is that the former doesn't require you to have K4PC installed on your computer
at all. If you don't use it... changes and updates to it can't affect you. This is obviously only a viable solution for those who own Kindle devices.
Same workflow, no ever-changing—potentially tool breaking, re-download causing—updates (recently).

Download via Amazon's webpage to your PC
(no K4PC... just because you're downloading the copy meant to be transferred via usb doesn't mean you actually have to
transfer it via usb
.
Import to calibre (plugin removes DRM with Kindle serial number).
Email to K3.
No USB cable involved.
