Error: startup error
Having trouble with Calibre. When I try to run I get a box with the startup error message and then a drop down box with this.

That error indicates that the ReadFile system call is failing, which is most likely a hardware error. Your disk is likely failing. Try running scandisk to check your drive for errors.

That did it. Must have been something wrong. Ran scandisk and afterword Calibre started right up. Thanks for the help guys.

If I were you, I'd look into replacing that disk as soon as possible.
