What model of Kindle? What version of firmware on the Kindle? What operating system on the computer? Does the Kindle mount on the computer without running calibre?

If it doesn't show up as a connected device outside of Calibre either, then try a different USB cable and a different USB port (preferably hosted by a different internal USB controller) on your computer.

Also, especially if you're running Windows, reboot your computer. The first thing to try for any Windows problem is "reboot the computer".
