That's because... wait for it... it couldn't mount it.
Why? I don't know.
You can try as normal vfat or ext4.

Tried another USB stick, it mounted as vfat. That didn't work either. Very weirdly as well, after it attempts to flash the zip the device no longer becomes bootable normally, just to recovery. I then have to flash the stock reovery, and do a factory reset for it to boot normally again.

I'll try reformatting the second USB stick that I have

So the USB drive had to be formatted to FAT32 for the update to work. Looks like the conversion process was successful. Thank you. I'm also a little curious - would the update.zip file have worked by copying to the SD card and installing it using the stock recovery?

Yes, devices with SD cards work fine.
But you have to push them in when it tells you, not before.
autoupdate can find any newly inserted disk.
I've used that on my Poke6.
