The permission denied error says that calibre attempted to access a location where the user does not have the necessary rights. Does it still happen if you disable McAfee? Or try restarting your computer in Safe Mode and then running calibre. Did McAfee or other programs install an update between installing calibre and when it broke?

If you look at results from using VirusTotal, McAfee is among the top 3 in false positives. In my testing about 2 years ago, McAfee did not find any infections that Microsoft's anti-virus did not also find and it found quite a few files that were listed as infected when neither Palo Alto, Microsoft nor Malwarebytes flagged the file.

The other functions that Norton and McAfee want to install should not be permitted near your computer, IMNSHO.

Someone on one of my Discord servers had some trouble a few years back with Sims 2 savegames. They're very touchy and if a certain file's locked in-use when you open the game it will corrupt the entire neighbourhood. Guess what McAFee kept doing?

The best AV to use for Windows (IMHO) is the one that comes bundles with Windows. It works well. I've not gotten any false positives. And it does not noticeably slow down the computer.

Yes, I do the same. I've gotten a few false positives but just exempt the folder.

MalwareBytes Anti-Malware is good run the manual scanner every so often, and run any suspicious executable through VirusTotal.

IMHO ANY product that makes it near impossible for the Admin to remove is in itself Malware. This is not to say that it should not protect itself from a drive by with a 'query image' (captcha like).

Come to think. You are correct.
Way back (W95 era) it in fact Launched the virus by checking for one in a ZIP. The did fix that flaw.
(I had already deloused my system by then)

But no, Not the 'Generic' yellow box, but another that still bears the originators name
