If that works it might mean the Installer has cleared the 'blockage' in the registry. You won't know that unless you uninstall from C: and reinstall on E:. Or you might want to let sleeping dogs lie and leave the calibre programs on the C: drive. There's no intrinsic advantage in having the calibre programs on the same drive as the calibre libraries… if anything the opposite is true.

I don't use Calibre Program on same 'place' as Calibre Library except on the VMs that have Calibre for testing. Sometimes a different partition, sometimes a different drive (i.e. programs on SSD and data on an HDD). The main issue is that both should be on local drives, no NAS/Shares/Cloud involved.

Also I close Calibre before doing an rsync based backup to server on the LAN. And any other programs that open files on the rsync folder/directory list.
