It may very well be the case that the Nova Pro has a glass substrate. Of this, I have no idea. However, the Nova Pro definitely has a glass screen.

I happen to have an original Note, a Note Pro and a Nova Pro. The screens on both the Note Pro and the Nova Pro are similar and they are both very different from the original Note which is very definitely a flexible screen.

But don't take my word for it...

Now, whether or not a screen protector would protect the glass screen on the Nova Pro from breaking, I haven't a clue. I don't feel the need for one myself. I simply keep it in a cover. That has provided all the protection I need.
