Have you experimented with using a different USB drive in the same USB port?

Have you experimented with plugging your USB D: drive into a different USB port?

BUT

I cannot change anything on D using Calibre.
I also run Foxit Reader for PDFs. If a PDF is on the D Drive, I can't save edits (such as adding bookmarks.) I can make a copy of the same PDF, move it to the C drive and save edits.
I ran Libre Office Writer, and cannot save to the D Drive.

In all cases I get this General Input/Output Error msg.

Some crapola related to Win10 is not allowing certain programs (including Calibre) to write to the D Drive.

It is not the drive. Chkdsk reports no errors. What can be causing this issue?
Check the permissions for drive D:. Also try running Calibre as administrator and see if that works. If Calibre works when run as administrator, then it is the permissions that need fixing.

A permissions problem wouldn't cause an I/O error message, and it wouldn't cause the USB D: drive to work at all times when not being used by Calibre. This error is only being generated when using Calibre. Also, whether being run as an Administrator or not wouldn't make a difference in the problem that results in an I/O error.

Not only Calibre. I can't save Libre Office files (.odt, .ods) to the D drive. I can't save changes to pdf files on the D drive (For example, Foxit Reader writes added bookmarks to the file).

No problems with any of these on the C Drive.

Something in Windows is restricting changes based on file types (or file extensions).

I can save and get no input/output error messages at all for .txt, .bat, .c, .cpp, .py, .json, and who know what else on the D drive. I have used non-MS products, not just notepad.

As you say, something in Windows is preventing...
Valid troubleshooting will be write to a different usb drive in that port, write to that drive in a different port, test with another pc....
I would leave calibre out of it until you have worked thru the above permutations.and, obviously, check or suspend AV settings.
sone AV may be blocking potentially executable file types so txt is allowed, epub being a type of zip is not...

Okay, I thought you said the problem only happens when using Calibre. Do you know how to check the permissions (under Properties/Security)? Make sure that Everyone or your user name or Administrators has access to everything, but I don't think that's gonna solve the problem, since you have no problem with some of the extensions.

Did you try the experiments I suggested with different USB drives and USB ports?

If it is a permissions issue, Google take ownership windows app. There are various free and easy to use utilities that make it easy to get full control of files or folders with a simple right click.

Also unlikely but check, is the bug linked to whether you are replacing {over writing} a file Vs creating a new one. If you don't have delete permission to remove the old file, it could manifest like this

I had a windows Version (1903) update mess up my internal SD card reader (I discovered it when I went to grap photos from my dashcam's SDcard). The fix was simple (once I knew I had a problem)
I opened the Device Manager: USB section and there was the error icon. I deleted the device: Scanned for Hardware changes and Presto: It installed properly this time

In subsequent posts you said other programs also had issues. I assumed you already power cycled the computer and the drive. If you still haven't tried running as administrator do it and report back your findings. Sounds like the calibre library may be read only. Don't assume how any program will report a permissions issue error. I/O error is as good as any when unable to write to a drive.

Here is a general description of an I/O error:

Causes of an IO Device Error
The hardware device that you are trying to access is damaged or defective. The hardware drivers are damaged or incompatible. There is a connection problem, such as a bad cable. The CD or DVD disk that you are trying to access is dirty or damaged.

I know what an I/O error is, I've been in I.T. for 4 decades. I also know that programmers may trap, handle, and report errors in countless ways that sometimes defy imagination. This means until you actually test the possibility all you have are assumptions.

Hell, I lost track of who we were trying to help. Without your 2 cents @Dae may have taken 2 seconds and then wrote a post saying it did or didn't work.

Good luck.
Ahem. deback is not the guy who asked for help.

but I agree that it would help if the OP (dae) actually replied to the many suggestion already given re testing/troubleshooting.
otherwise - why are we bothering

update. I re read the whole saga.
I confess I thought we were talking about a calibre library on a USB stick but in one post the OP says its a multi-terrabyte drive. ( if that's a self powered one then previous speculation about getting enough current from the USB port for sustained RW stays on the usual suspects list)

I also know that when I have moved fixed drives to new PCs I have encountered ownership issues. windows thinks the drive is "owned" by the other PC so it blocks some operations when attempted on the new PC. the easy fix for that is a take ownership utility or a registry hack to get full ownership of that drive for the current user on the current PC

all the other ( ignored?) troubleshooting suggestions of trying various permutations of different PC, different port, different usb drive remain valid and will help , if OP can be bothered. ( & if he can't its not going to get solved )

that's me finished here, unless he bothers to report back - its been 2 days and counting since OPs last post.
