Okay, I just converted my EPUB 2 book to an EPUB 3, and now ADE won't open it at all. "Errors were encountered in this item". It's a valid EPUB 3, I've checked it both with the command-line epubcheck and at validator.idpf.org. How can I coax a more verbose error out of ADE? (It still works in Readium.)

In that case your book might contain an HTML/CSS syntax error that epubcheck doesn't check for or it's an ADE bug. Can you post an excerpt of the epub3 version of your book that also can't be opened with ADE or preferably the complete book obfuscated with Borkify ePUB?

I don't think you can. Try opening it with other ePub3 apps. E.g. Azardi or Gitden Reader. If it opens with them, it's most likely an ADE 4.5.1 bug. (Adobe has a long tradition of introducing annoying bugs with each new ADE generation.)

Okay, must have been something odd with that ADE install, because I tried the same book with ADE on another machine and it worked fine. So, back to the original subject: it mostly works. It doesn't let you move through the pages forwards, but once you're at the end you can go backwards through all the pages linearly. Which is annoying but I can live with it.

If you limit yourself to ePub3 apps, you might want to consider Javascript solutions, because they'll give you greater control over link handling.
For example, the first topic of the test epub that I linked to uses LocalStorage and JavaScript to hide/unhide text.

I also created two simple JavaScript epub solutions that you might be able to adapt to your needs. You can find them in the Some tricks with ePub 2.x (wanted) thread. (The first one stores the id of the calling href and the second one dynamically loads text from a separate file.)

ADE 4.5/RMSDK 12.5 does support LocalStorage, but only in ePub3 books.
You can test this yourself with this ePub3 book.

I just successfully tested my test epub3 book with the current Windows 10, OS X (Yosemite), Android and iOS versions of ADE 4.5.x.

However, I used a slightly more recent OS X ADE version than you. (I used the version from the Adobe download page.

(You can tell that LocalStorage works, if you tap/click the Off button on the first page of my test book and the interlinear text stays hidden, when you close the book and re-open it later.)

Meanwhile I tested also on a Os 10.11.3 and it is not working... I put your ebook on ADE, I open it, I see the blue and black text, I click OFF to see only black text (not blue), I leave the ebook and go back to Library, I open again the ebook and I see the blue and black text again.
The problem is not in your ebook, my test ebooks (EPUB3) have the same problem. I tested this on different Mac computers.

It looks like the OS X version of ADE deletes the LocalStorage file when a book is closed, which is rather strange, because it doesn't do this on the other platforms. IMHO, this is a an ADE OS X bug.

However, as long as the book is open, it's possible to read and write LocalStorage data, which is, of course, of limited use.

Actually I'm really sad about localstorage support. Localstorage could be a real BIG change in ebook design. But the ebook readers support is poor: ADE do not support it on OSX (or, only until the ebook is open, that is a too limited use), Ibooks seems to support it... now. But in the past Apple turned on/off the localstorage support: some year supported it, some year stopped the support. On my Android ebook reader (Onyx M96) Gitden Reader supports it... until you turn off/on the device. Sigh.
