Pressure DOES matter

I've tried the light tap and hold and the page turns for me. If I exert SLIGHTLY more pressure and hold, the definition turns up.

A light tap and moving the finger activates the highlight.

Maybe the OP should try my method before giving up altogether.

There may be a layer underneath which needs contact to activate the dictionary. Its also possible that his unit is defective.

There either is a contact or there is not. The touchscreen cannot detect the amount of pressure you're exerting on the screen, but simply the presence or absence of a contact.

But yes, you certainly need to touch the screen firmly to ensure that the contact is properly registered.

Sometimes this problem (Kindle not recognizing holding, but interpreting it as tapping) is due to slowness of the system. Maybe you loaded too many books at once? Kindle may be indexing them and, so, the system is slow for a time. If not, try to simple reset the Kindle (menu > config > menu > restart), so it can breath.
