It's alleged to support them--we don't know yet. And what we're discussing is how it would work across all Kindle devices, on the Mobi front, as the other devices patently (no pun intended) don't have pop-up footnotes--and 95% of ePUB readers don't.

We don't even know if the functionality will be provided to self-pubs--just like Audio/Video; it can be done, it just cannot be published by a self-publisher. We'll have to wait and see.

Sounds like it might be just some sort of heuristic processing of links in existing ebooks. At least I don't think there's been an updated edition of Infinite Jest released to showcase the new footnote feature or anything.

Reasonable assumption. I looked how the first footnote was implemented (from an old epub converted by Calibre, not the Amazon azw3).
