No. AWZ3 is merely a file extension, and can contain any type of file. It so happens that "The tools that shall remain nameless" give a KF8 file a ".AWZ3" extension, but that's just what those tools do; there's no reason that a KF8 file has to have a ".AZW3" extension and there's no reason that a file with a ".AZW3" extension should contain a KF8 file. If you create a KF8 file with "KindleGen", for example, it will have a ".mobi" extension.

It's as well to separate the two things. KF8 is not AZW3 and AZW3 is not KF8.

I mean when you convert a file in Calibre AZW3 is KF8 format. You can choose to switch to the new mobi format in the conversation window but the OP was asking about Calibre and which format to use. It's just easier to use AZW3 to keep the formats separate and not become confused.
