Try turning off bluetooth, notifications and location services in the Settings app.
I had a similar problem with my 3rd Gen iTouch draining overnight after I upgraded to iOS 4, and this seemed to fix it.

I checked my settings and both bluetooth and location services were already turned off. Notifications was on so I turned that off but not sure that will help much. I'm also already upgraded to 4.1 software. I think it is the wifi that really drains the battery.

Well, I finally got a charger for my Itouch. My sister bought me a Gomadic tip exchange charger for my Sony Reader for my birthday so I just had to order a tip for the Itouch. Works great! Now I only have to deal with one cord. The Sony doesn't require charging that often so no problems sharing the cord. Got a charger for my car too. I'm good to go. No more firing up Itunes just to charge the podster.
