Is it possible to have a column which shows a calculation?

I've got two existing columns which contain dates. A start reading date and an end reading date.

Is it possible to have a third column which would show a value in days between the start and end date so I can see how long it's taken me to read a book roughly?

Sure, you can use the days_between() function in General Program Mode in a custom column built from other columns.

days_between(
    raw_field('#end_date'),
    raw_field('#start_date')
