Pulling ISBN after a book has been added

Is there a way to pull the ISBN from the book after it's been added to
a library ? Or do I have to add a regular expression then re-add the book(s) ?

Neither, as far as I recall calibre doesn't have the ability to extract the ISBN from the book itself only from the metadata that may have been in the book.
