Jay: excoriate ... I think it's pretty self-explanatory, actually: it means just what it sounds like it means.

Nay: Oh there are so many, but I'll go with "juxtapose" when the speaker means "diametrically opposed" -- they're not even remotely related!

Qualifies as both: jay, it's the longest word in the English language, and requires multiple readings to parse; nay, it's a real pain in the sitter-downer to spell ... and requires multiple readings to parse.
