Can Calibre work with 2 ereaders alternatively?

Hi there, I am new to this forum. My wife reads from a Kobo Glo and I read from an iPad. We would like to have our ebooks organised in one laptop using one ebook manager. So far I was able to sign up only one reader with Calibre. Is there a way to work with 2 readers?

Yes, calibre can handle any number of devices as long as you don't connect more than one at a time.

Calibre doesn't really "sign up" devices. I suppose you mean you the setup wizard and selected one of these devices. Doing that really just sets a the maximum image for when you convert a book. You can always choose a different size when converting, but, I recommend just using the "tablet" option. This will mean the images are not resized and the device will do whatever is needed. For both the Glo and an iPad, this will work.

You might also want to use the Reading List plugin. This allows you to control for each device which books get sent to it.

At this point in time, I don't believe Calibre can directly interface with the iPad via USB.

OP would need to go wireless (content server or Calibre Companion).
