PDF to Mobi makes multi-columns

Just downloaded 64-bit Calibre and tried to convert a PDF.

The Mobi file has columns. The original PDF does not.

Is there a setting I need to change?

I tried converting your PDF file, I see no columns in the MOBI. If you mean that the lines are short, that is because they are short in the PDF as well.

THere are no columns in the file, indeed the MOBI format is not capable of having columns. If youa re seeing columns they are being generated by whatever software you are using to view the MOBI
