Are you talking about the Kobo Desktop app (which you don't need at all - just get rid of it or ignore it), or the home screen on your Aura HD? If the latter, just long-press on the tile you don't want, and choose Dismiss Tile.

Firstly, the main screen of the Kobo devices is usually called the "Home page" or "Home screen". Your thread title confused me as "Kobo Desktop" usually refers to the Kobo desktop application for Windows or Mac.

Strictly, you don't need to. The home page is dynamic. As you do things, they appear on the home page as a tile. If you don't use something that is on the home page, it gets pushed off. So, open nine books, and they will replace all the tiles on the home page and all the others (except one) will disappear.

If you want to remove a tile, you do a long press on the tile until the menu is displayed. One of the options on the menu is "Dismiss tile". Press this and the tile is removed.

Tthere are two special tiles. The first tile is always your current book. It shows the book that was most recently opened. This tile can't be dismissed, but you can dismiss the book and the next most recently opened will take its place. Under the current book tile will be the "Sync" tile. This can't be dismissed and if you long-press on it, no menu is displayed.

You can do some of this from calibre. My Kobo Utilities plugin has a "Dismiss tiles" function. This allows you to select the types of tiles and have them removed from the home page. The function can also create a database trigger that attempts to prevent the tiles from coming back. It doesn't work perfectly, but does prevent most of the tiles from coming back.
