The easiest way would be to sort that column, and then highlight the null entries and set them to whatever value you like, using bulk metadata edit.

is there a means to make it Automatically (command code, pithon....access to the db)

not for each. Only once for all 500. That is why it's called bulk update.

Like he said, sort on the field, (shift-)select all the ones without any value...

No! Not 500 times

Select the Group
Tap 'E' (Bulk Metadata edit)
Locate the field on the form: Fill in the new value: Save
1 operation on existing books.

But instead of modifying the DATA, make and assign an Icon for when the field is blank (undefined) . Preferences: Look and Feel: Column Icons

Or just sort on that field. Click the column header. All the books with no localisation will then be grouped together. And you can select them all simply by clicking on the first, and shift-click on the last book.

Additionally you can add the column and it's values on the sidebar [like the authors] and that will help you to select each value with one click. [Manage User Categories, left bottom corner]

This will not set values but will give a quicker access to the books marked with the values. My example is the READ and the following three categories.
