Memory Card Slots

Does anyone know if you can use both of the memory card slots (MS Duo & SD) in the PRS-600 at the same time?

Yes, I do believe that you can, though (depending on the size and amount of content on said cards) your initial start-up times may increase dramatically.

Personally, I'd stick with MS Pro Duo cards, as they tend to be a bit faster overall.

Yes, definitely possible. Has been possible on Sony 505 and Sony 700 already.
Sony even has implemented a workaround into Adobe Digital Editions. Thanks to Sony's proprietary driver, in Adobe Digital Editions you see: Internal Memory, SD card AND Memory stick. So you can drop your books wherever you want to. Cybook Opus for example only does show the internal memory, not the SD card.

Startup time only increases, if you've made changes since the last start-up. If the index already exists, the number of books doesn't matter.
