If I have purchased a book via the Amazon site, am I able to run the file through Calibre to remove the DRM so I can let my brother read the same book?

DiapDealer is right in that Calibre itself does not remove DRM. But becoming friends with Apprentice Alf helps (Google is your friend too)

I have a plug in that allows me to remove drm and time restrictions from books borrowed from libraries, so I can run them on my Kindle.

I wasn't sure if there was a similar one for .mobi books

We cannot discuss DRM removal here. Calibre itself has no facilities for DRM removal.

Note that Amazon provide a perfectly legal method for you to share your eBooks with your brother; simply register his Kindle to the same account as your own.
