Should depend on the format. Calibre are capable of editing epub and azw3. You can open it there and change the metadata that way.

I opened an ePub file from my Dropbox to Calibre and there seemed like nothing to change. If I make changes on Calibre to the ePub file pulled from Dropbox, does it automatically make the changes to the file in Dropbox? I don't see save function.

Bingo
Books WITHIN the Calibre Library do not Get updated with new metadata/covers automatically.

You need to Polish or use Modify Epub
Or
Use the Book Editor to add/change a internal file

Notice I did not List convert as a preferred choice. That is because it may introduce other changes. IMHO more of a Last resort l
