You are not the first person who has reported that Windows 10 is not reporting their ereader as a removable drive.

Basically, it is a Windows bug.
Reinstalling Windows from scratch has been suggested as a solution -- it seems the problem generally turns up on computers that were upgraded from Windows 7/8/8.1 and not cleanly reinstalld.
