You can save yourself a lot of search and replace actions by using
Calibre with Kiwidude's
Modify Epub plugin. Simply select "Smarten Punctuation" and let the plugin do its magic.
Note that the heuristic algorithm is pretty good, but not 100% perfect.
If you don't want to use Calibre you can also test the "Smarten Punctuation" feature with DiapDealer's
SmartyPants Python wrapper. (Both use the same python script.)
