Thank you. I did all of what you said and still no success. So I decided it must be a problem with the Nook itself and called their support line. After an hour and a half on the phone, I finally found the solution. Perhaps others will have the same problem, so I will tell you what needs to be done. When you plug your Nook 10.1" tablet into the computer it assumes that you want to charge it only. You have to tell it to copy files instead.pull down a menu by stroking down from the top of your Nook. The very bottom item on this menu says "Android System - USB charging this device. Tap on this line. Now you see the line expand and it reads thus: "USB charging this device. Tap for more options." Tapping on this will bring the following menu: Use USB to charge this device, transfer files, and transfer Photo (PTP). Choose transfer files. Once you do this, Calibre sees the Nook. You have to do this EVERY TIME you plug into your computer because it always defaults to "charge this device."
