No. All you can do is, using the Connect software on a PC, delete the file from the memory card and then reload it into main memory (or vice versa, of course). No way to move or delete from from the Reader itself.

Speed-wise, there's no difference. Putting books in main memory gives you better battery life, and given the fact that books in main memory are compressed (those on a memory card aren't), you can store a LOT of books (100+ normal sized novels) in main memory.
