This indicates that the book does not have its language set to "English" in its metadata. Look at the OPF file inside the ePub - it should say:

<dc:language>en</dc:language>

If you've converted it with a non-current version of Calibre, it probably says:

<dc:language>UND</dc:language>

and that prevents the dictionary from working.
