This "fast refreshing, colorful" screen is, for a majority of people (definitely not all!) not suitable for extended text reading, you yourself give the two points that alone justify a reader. Others are heat and battery service life/cost. I see the relationship between a dedicated device like a reader and the current crop of multifunction devices as the same as between a camera phone and an SLR. Sure, you can make a few snap shots with your phone, but if you are serious about photography?
Besides, the $499.- is not really a serious entry point. 16GB storage for such a device? So we are really talking $799.-. I would love a multifunction device that can also replace a real computer, with a screen that is easy on the eyes and a battery that can last a few days. This gigantic ipod still cannot even run background apps! So for me, I see no place for it, nothing it can do that I can't do much better with some other device I already have.
If you love a dedicated browsing device, multimedia player and are not satisfied with a phone with a big screen -- then go for the ipad. I just don't think it is worth the money compared to a good netbook and can't satisfy me as a reader. For those people that read comics and complicated PDFs, maybe.
