I pronounce it liz-use. With a rather soft "y" sound on the "use."

If that's wrong, it doesn't need to be fixed; it's not like I'm likely to ever say it aloud.

I like the idea of the word, but don't think it'll catch on in the US; it doesn't connect to anything we think of as meaning "book." I suspect that, in the long run, ereader is going to be the word of choice, and eReader-the-program/filetype is going to lose their trademark for not enforcing it on the hundreds of blogs that discuss ebooks.
