If you're using ADE, then yes, calibre and ADE must be on the same computer. Alternatively you can use the ACSM plugin, you won't need ADE then.

And copying epubs from the Pocketbook app is not the way to deDRM them. You must either use ADE or the ACSM plugin. In the latter case you must import the acsm file into calibre, not the epub.
