Debug mode (Calibre Preferences dropdown -> Restart in debug) will give more logging.
Yeah, something's gone wrong with your epub, or something is preventing FFF from reading it.
If it's just the one book, I'd restore from backup or just delete the epub and download new.
If it's happening to lots of books, I'd check if you're out of space (should be obvious from all the other issues if that's happened). Other options include failing drive, changed file permissions and over-zealous 'protection' software.
