FWIW, Apple Books does let you adjust line, character, and word spacing ('Customize' in Aa menu).

You can do some color manipulation with Accessibility Display options, but at most you might be able to give what would be a white background or text some tint. My preference is some Dark theme on pretty much any backlit screen.
