No, the Content server supports Web browser and also on non-Kindle, the OPDS system (Pocketbook app on Android etc). I forget how KOReader on a Jail Broken Kindle accesses the Calibre Content Server.

The Content Server is included in Calibre.

What's the problem with USB?

Also KFX output from Calibre, last time I looked, actually uses an Amazon program installed on your PC, so Calibre on Linux with USB to Kindle only reads KFX (and only if you have the suitable plugins and Kindle ID for the ereader, which can be copy/pasted from the Amazon "My Devices").
