I use Defender too.
I'll keep on checking some books next days, and according to Calibre behaviour, I will maybe uninstall Calibre and install it again.
But it seems to me more and more that problem is not into
C:\Program Files
but inside C:\users\MyUserName\AppData
In that case, uninstalling and reinstalling wouldn't change anything.
