The driveinfo.calibre and metadata.calibre files and folder .kobo have a date modified today.
Have you tried moving the book file to a different location on your Kobo (since you mentioned the .kobo directory, please tell us that you don't store books in it). For this one book for testing, you could even use the root of the exposed partition.

Remember if the book is not imported into the Kobo database, calibre will not see it as being on the device other than immediately after sending it.
